The aim of this HDR is to review the advances and work about phototransformable fluorescent proteins (PTFPs) Virgile Adam has contributed to, since his doctoral thesis on this topic nearly 15 years ago. It is articulated around the various photophysical phenomena of PTFPs: reversible photoswitching between on and off states, irreversible photoconversion from a color to another and biphotochromic fluorescent proteins, photophysical constraints on the use of PTFPs in super-resolution PALM microscopy and the emerging field of cryogenic PALM.
