Actin plays a key role in various cellular processes involving the assembly and disassembly of highly ordered, dynamic structures. However, little is known about how actin and its interactors are organized within cells to generate the nanonewton forces that sustain these processes. In recent years, cryo-electron tomography (cryo-ET) has become the method of choice for structural analysis of the cell interior at a molecular level. In this talk, I will present our past and ongoing cryo-ET work into various cellular actin structures and demonstrates how this work has provided new insights into in situ assembly at the actin cytoskeleton-membrane interface.
