Molecules of innate immunity and pathologies

Several proteins of our research topics are also implicated in different pathologies. In this context, our goal is to decipher the underlying molecular mechanisms that lead to physiological disorders. All of these investigations are made in collaboration with other research teams.

Functional study of gut microbiota serpins as anti-inflammatory agents

Serpins (Serine Protease Inhibitors) derived from the human gastrointestinal tract (GIT) microbiota have been recently discovered as being under-represented in Inflammatory Bowel Diseases (IBD) patients. These diseases, characterized by an over-expression of human serine proteases found in the GIT reveal that the serine protease regulation is hugely disrupted in case of IBD. The goal of the project is to characterize bacterial protease inhibitors (Serpins) from microbiota of healthy patients, succeptible to control the gut inflammation in case of IBD.

Functional implications of the alterations of complement proteases C1s and C1r identified in Ehlers-Danlos periodontal syndrome patients

A recent linkage between the Ehlers-Danlos periodontal syndrome and heterozygous mutations in C1s and C1r genes has been reported recently. The objective of this collaborative project is to clarify the clinical symptoms and the molecular mechanisms that are responsible for this rare disorder using a multidisciplinary approach. We will analyze the structural and functional impact of these mutations in order to relate them to their clinical consequences.

Role of anti-ficolin auto-antibodies in the pathogenesis of Lupus nephritis

The dosage of auto-antibodies against ficolin-2 and ficolin-3 in a cohort of patients with a disseminated erythematosous lupus allowed us to show a correlation with clinical manifestations, particularly in the kidney (active nephritis). We aim at understanding the mechanisms of action of these auto-antibodies on the function of ficolins, at a molecular and cellular level.
